HPLC法测定苦参中苦参碱的含量*

摘 要 目的：建立苦参中苦参碱含量的测定方法。方法：采用HPLC法，色谱柱为Kromasil C18柱(250mm×4.6mm，5μm)，流动相为甲醇-水-二乙胺(55∶45∶0.02)，检测波长208 nm，柱温40℃；流速1.0ml.min-1；进样量：10μl。结果：苦参碱在0.05～1.25μg 范围内具有良好的线性关系（r＝0.9999），平均回收率97.62 %，RSD为1.15%（n＝6）。结论：本方法灵敏、准确、简便快速，可用于苦参药材的质量控制。

DETERMINATION OF MATRINE IN RADIX SOPHORAE FLAVESCENTIS BY HPLC

Abstract Objective：To establish a method for the determination of matrine in Radix Sophorae Flavescentis．Methods：A Kromasil C18 column（250mm×4.6mm，5μm）with mobile phase consisting of methanol-water-diethylamine(55∶45∶0.02) was adopted，and the detecting wavelength was set at 208 nm. The column temperature was 40℃. The flow rate was 1.0 ml?min-1，and the sample size was 10μl．Results：The calibration curve showed good linearity over the range of 0.05～1.25μg（r＝0.9999）. The average recovery was 97.62 % with RSD 1.15%（n＝6）. Conclusion：The method is sensitive，accurate，rapid，and is suitable for the quality control of Radix Sophorae Flavescentis．
