肾上腺髓质素对大鼠心肌成纤维细胞增殖的影响*

摘 要 目的:研究肾上腺髓质素(ADM)对幼年大鼠心肌成纤维细胞(FBC)增殖的影响,并探讨其可能的作用受体。方法:用差速贴壁分离法获得心肌成纤维细胞为材料,用免疫组化的方法鉴定细胞,并绘制生长曲线。用MTT比色法测定细胞增殖。结果:①ADM对基础状态下的FBC增殖无明显抑制作用,但能呈浓度依赖性地抑制AngⅡ刺激的FBC增殖。②CGRP8-37和ADM22-52均能部分拮抗ADM对AngⅡ的抑制作用,而二者合用几乎能完全拮抗。CGRP8-37对ADM的拮抗作用强于ADM22-52。结论:①ADM具有浓度(10-8M~10-5M)依赖性地抑制AngⅡ介导的FBC增殖的作用。②心肌成纤维细胞上可能同时存在CGRP受体和ADM特异性受体,但其作用可能更多通过CGRP受体。

RAT CARDIAC FIBROBLAST

Abstract Objective:To investigate the role of adrenomedullin(ADM) in the proliferation of rat cardiac fibroblast,and to probe into its possible receptor.Methods: Neonatal rat cardiac fibroblast(FBS) were isolated and cultured in vitro,FBS proliferation were determined by MTT assay. Results:①ADM had no significant effect on the proliferation of FBS in basal state, but could inhibit the FBC growth stimulated by AngⅡ in a concentration-dependent manner.②Both CGRP8-37 and ADM22-52 could markedly inhibit the antagonistic action of ADM attenuating on the cell growth induced by AngⅡ, but these actions were not complete. If the two receptor antagonists were incubated together, they could completely block the role of ADM. It was shown that the action of CGRP8-37 was stronger than that of ADM22-52. Conclusion:①ADM can inhibit the FBC proliferation induced by AngⅡ in a concentration-dependent manner in vitro.②There may be two kinds of receptors in FBC. One is CGRP receptor, another is specific ADM receptor. ADM may act more through the CGRP receptor.
