肺癌细胞株c-erbB-2基因与放疗抵抗的相关关系*

摘 要 目的:探讨人类表皮生长因子受体-2(c-erbB-2)基因与非小细胞肺癌细胞株放疗抵抗的关系。方法:采用RT-PCR测定肺腺癌(CALU-3和A549)和鳞癌NCL-H520细胞株的c-erbB-2
mRNA水平;采用流式细胞术检测CALU-3、A549和NCL-H520细胞株的c-erbB-2蛋白水平;绘制CALU-3、A549和NCL-H520细胞株的存活曲线。结果:肺腺癌CALU-3细胞株的c-erbB-2
mRNA水平均高于腺癌A549和鳞癌NCL-H520细胞株,差异有统计学意义(均P<0.05),而A549和NCL-H520细胞株之间差异无统计学意义(P>0.05);CALU-3细胞株c-erbB-2蛋白的阳性率均高于A549和NCL-H520细胞株,差异有统计学意义(均P<0.05),而A549和NCL-H520细胞株之间差异无统计学意义(P>0.05)。存活曲线显示,
CALU-3、A549和NCL-H520细胞株D0值依次为1.18 Gy±0.05 Gy、1.67 Gy±0.13Gy和1.50
Gy±0.26Gy,细胞株间差异无统计学意义(F=0.287,P> 0.05),Dq值分别为3.98 Gy±0.03Gy、2.43
Gy±0.47Gy和1.32 Gy±0.08 Gy (F=61.670,P<0.05),其中CALU-3和A549、A549和NCL-
H520、CALU-3和NCL-H520细胞株之间差异有统计学意义(P分别为<0.05、<0.05、<0.01)。结论:肺腺癌CALU-3细胞株的c-erbB-2基因在mRNA和蛋白水平均高于腺癌A549和鳞癌NCL-H520细胞株;三种细胞株中,CALU-3对γ射线最不敏感,
NCL-H520最敏感;推测c-erbB-2基因高表达可能与肺腺癌CALU-3细胞株放疗抵抗有关。

RELATIONSHIP BETWEEN C-ERBB-2 GENE OF
LUNG CANCER CELL LINES AND THE RESISTANCE TO RADIATION

Abstract Objective:To investigate the relationship
between epidermal growth factor receptor-2 (c-erbB-2) of non-small
cell lung cancer cell lines and its resistance to radiation.Methods:
The c-erbB-2 mRNA level of the human lung adenocarcinoma cell
lines CALU-3 and A549 and squamous cell carcinoma NCL-H520 cell
line was assayed with RT-PCR;the c-erbB-2 protein expression
of the three cell lines was measured with flow cytometry;and
the parameters of survival curves of the cell lines was measured.Results:
The c-erbB-2 mRNA relative expression in CALU-3 cell line was
0.77±0.07,significantly higher than those in the cell lines
A549(0.65±0.05, P<0.05) and NCL-H520(0.62±0.05,P<0.05).
There was unconspicuous statistical significance in the difference
between A549 and NCL-H520 cell line(P>0.05). The c-erbB-2
protein positive expression rate was 33.78%±13.45% in the cell
line CALU-3,significantly higher than that in the cell lines
A549(15.60%±6.37%,P<0.05) and NCL-H520(4.22%±0.10%, P<0.05).There
was no significant difference between A549 and NCL-H520 cell
line (P>0.05). The D0 of the three cell lines was similar(F=0.287,P>0.05),while
the Dq of the three cell lines was significantly different(F=61.670,P<0.05).
The Dq of CALU-3 cell line was 3.98 Gy±0.03 Gy, higher than
that of A549 cell line (2.43 Gy±0.47 Gy,P<0.05) and NCL-H520
cell line (1.32 Gy±0.08 Gy,P<0.01);in addition, the Dq of
the other two cell lines was also significantly different(P<0.05).Conclusions:The
level of c-erbB-2 mRNA and protein in CALU-3 cell line was higher
than that in A549 and NCL-H520 cell line,respectively. Among
the three cell lines, CALU-3 cell line was more resistant to
the γ-rays than the others, and NCL-H520 cell line was the most
sensitive. The high expression of c-erbB-2 gene is probably
related to the radioresistance of CALU-3 cell line.
