小鼠尾尖成纤维细胞的分离与培养

摘 要 目的：建立一种简单的鼠尾成纤维细胞分离、培养的方法。方法：剪取小鼠尾尖，采用组织块培养法，用 DMEM(含10％胎牛血清)培养基进行培养，并通过传代得到纯化的成纤维细胞。结果：原代培养的成纤维细胞特征典型，并通过传代培养得到了纯化的成纤维细胞。结论：采用组织块培养法成功地培养了成纤维细胞，方法简单，为动物细胞核移植、转基因等提供核供体材料奠定了良好基础。

ISOLATION AND IN VITRO CULTURE OF MOUSE TAIL-TIP FIBROBLASTS

Abstract Objective： To establish a simple method for isolation and culture of mouse tail-tip fibroblasts . Methods：The mouse tail-tip tissue was cut into small pieces and cultured in DMEM(10% FBS) at 37℃ under 5％CO2 ;the cells were purified by succeeding generation-to-generation culturing . Results： The cultured cells showed typical shape and features of fibroblast cell ,which were successfully dissociated and purified．Conclusion:This technique is suitable for culturing the fibroblast cells， which might be expected to provide donor nucleic for future nucleus transplant and transgenic researches．
