摘 要 目的:研究丹参酮ⅡA磺酸钠(sodium tanshinoneⅡ-A sulfonate,DS-201)对单侧输尿管梗阻(UUO)病人肾间质纤维化来源的成纤维细胞(hRIFs)体外增殖及细胞周期素D1(cyclin
D1)蛋白表达的影响,探讨该药治疗肾间质纤维化的作用机制。方法:体外培养鉴定hRIFs;用四甲基偶氮唑(MTT)法检测对照组与不同DS-201浓度组hRIFs的增殖活性;用免疫细胞化学SABC法和图像分析技术检测对照组与不同DS-201浓度组hRIFs
cyclin D1基因的表达。结果:随药物浓度的升高和作用时间的延长,抑制率逐渐升高,抑制作用逐渐增强,呈剂量依赖性和时间依赖性;用药组阳性细胞的光密度值在第3、5、7、9天显著低于对照组(P<0.05,P<0.01﹚,第1天各组之间无统计学差异(P>0.05)。结论:①
DS-201对UUO病人hRIFs体外增殖有显著抑制作用,可能是其治疗肾间质纤维化的机制之一;② DS-201抑制UUO病人hRIFs体外增殖可能是通过抑制细胞中cyclin
D1基因表达,延长细胞周期实现的。

Abstract Objective: To discuss the effect of
sodium tanshinone II-A sulfonate (DS-201)on human renal interstitial
fibrocytes(hRIFS) of UUO and the expression of cyclin D1 and
approach the possible mechanism. Methods:HRIFs were assessed
through culture in vitro;the propagate activity of HRIFs of
control group and different concentration group of DS-201 was
detected by MIT method.The expression of cyclin D1 were analyzed
with immune cytochemistry SABC and the image-analytical technique.Results:With
the increase of drug concentration and action time,the inhibition
ratio and strength increased,and it was dose-dependent and time-dependent.
The average optical density of positive cell of drug-using group
was much less than control group on the third ,fifth, seventh,
and ninth day(P<0.05,P<0.01).There were no significant differences
between groups on 1st day (P>0.05). Conclusions: ①DS-201 can
significantly suppress in vitro generation of hRIFs,this may
be one of its mechanisms to treat renal interstitial fibrosis.
②DS-201 suppress in vitro generation of hRIFs is possibly through
inhibiting the expression of cyclin D1 and extending the cell
cycle.
